## What the FY2027 request buys

**FY2027 planned work.** - Generalize computational models by incorporating available data from public protein databases. - Incorporate machine learning methods with physics-based models to provide speed and accuracy of function prediction. - Evaluate the optimized model performance in capability demonstrations to predict molecular function from folding, binding and cooperativity. - Validate the developed computational models on select datasets to assess overall model performance in drug-resistance, antibody and enzyme design.

**FY2026 to FY2027 change.** The FY 2027 increase reflects testing and evaluation efforts to integrate program models and perform additional capability demonstrations.

**FY2026 plans — current year.** - Design rapid computational models on a small subset of protein families to incorporate the dynamics and function of proteins. - Evaluate models for the correlation of dynamics and function of known protein sequences to assess model extensibility. - Evaluate computational model performance in a capability demonstration to predict the unknown molecular function of novel sequences.
