## Summary

This activity requests $18.2M in FY2027, 31% of project MT3, down 21% on FY2026. The R-2A exhibit describes it across FY2025–FY2027, including what the FY2027 money is planned to buy.

## What the FY2027 request buys

**FY2027 planned work.** - Execute efficacy testing across diverse pathogens and chemical exposures to address respiratory distress, neurocognitive impairment, and vascular dysfunction. - Perform immune modulation effectiveness studies to reduce symptoms induced by viral pathogens. - Validation of efficacy and safety platforms for human-directed therapies to evaluate unintended effects and enable robust risk/benefit determinations. - Advance at least two investigational new drug candidates with supporting biodefense data into later-stage development. - Continue development of comprehensive searchable repository for repurposed pharmaceutical candidates. - Continue development of well-defined testing models for therapeutic candidate evaluation. - Continue anti-viral layered medical defense concept to broaden countermeasure protection.

**FY2026 to FY2027 change.** Decrease due to completion of FY 2026 clinical trials and divestments in toxin countermeasures.

## Before the request year

**FY2026 plans — current year.** - Continue to execute proof of concept efficacy studies for layered medical defense. - Continue to execute efficacy studies for down-selected lead drug candidates. - Continue development of well-characterized models for testing of advanced therapeutic candidates. - Complete studies to transition one broad spectrum therapeutic candidate to the Agent Directed Therapeutic Program. - Complete studies to transition one host-directed therapeutic candidate to the Host Directed Therapeutic Program. - Execute studies to transition broad spectrum antimicrobial data package to the Rapid Acquisition and Investigation of Drugs for Repurposing Program.

**FY2025 accomplishments.** - Transitioned 2 broad-spectrum, agent-directed therapies from early research into advanced development phases. - Progressed development of non-ionic surfactant vesicles and nanolipid particles as platform delivery technologies to minimize dosing requirements and mitigate adverse side-effects. - Continued development of 3 leading host-directed therapeutic candidates for alleviation of respiratory distress resulting from viral infection. - Continued characterization of alternative testing models to facilitate advanced development of broad-spectrum antivirals through FDA regulatory approval.

## Funding

| Fiscal Year | Estimate Type | Amount ($M) |
|---|---|---|
| FY2025 | Actual | 21.1 |
| FY2026 | Enacted | 22.9 |
| FY2027 | Request | 18.2 |

> Prior, current and budget year only — an R-2A activity carries no five-year plan. It sums exactly into its project in the request year and not necessarily in any other.
