What the FY2027 request buys

Verbatim from the R-2A exhibit for project MT3 of PE 0603384BP. This is the budget justification's own description of work that has not happened yet — the one thing no other level of the budget carries.

FY2027 planned work

- Finalize development of comprehensive searchable repository for repurposed drug candidates containing supporting data for biothreat pathogen applications. - Advance broad spectrum, repurposed pharmaceutical candidates with biodefense evidence toward later-stage development or clinical practice guideline establishment. - Initiate development of broad spectrum nanoparticle delivery platform for therapeutic encapsulation and formulation. - Initiate development of human-relevant multi-organ system incorporating both innate and adaptive immune components.

Before the request year

FY2025–FY2026: what came before

Prior-year accomplishments and current-year plans from the same exhibit. Context for the FY2027 plan, not a series — an activity partitions its project exactly in the request year, but can under-cover it in earlier years.

FY2026 plans — current year

-Continue the development of 5-10 repurpose broad spectrum capabilities. -Continue to create and sustain curated, searchable databases of molecules with toxicity, drug development and efficacy data to decrease timeline in MCM fielding. -Continue to develop host-directed pathogen agnostic therapies directed at mitigation of presentation of disease. -Continue to develop testing platform to examine off-target effects and/or unintended consequences of therapies directed against host targets.

FY2025 accomplishments

- Continued studies demonstrating the broad-spectrum efficacy of an FDA-approved muscle weakness treatment for reversing toxin-induced muscular paralysis. - Finalized studies supporting the application of an FDA-approved clotting disorder treatment as a therapeutic intervention for vascular leakage in hemorrhagic fevers. - Initiated studies generating biodefense data for a viral entry inhibitor currently in Phase 2 clinical development. - Concluded efficacy assessments of approved antimicrobials against biothreat pathogens to guide clinical practice recommendations.

Three years, and no five-year plan

An R-2A activity publishes the prior year, the current year and the budget year. The FYDP outyears exist at project and program-element level and are deliberately absent here rather than inferred. Estimate types are colored and never summed into one figure.

This activity is 34% of project MT3's FY2027 request and 6.4% of PE 0603384BP's. In the request year the activities under a project sum to it exactly; in the current year they under-cover it in about 9% of cases, so an activity's delta can legitimately exceed its parent's and the two must not be compared row to row.
